Abstract

MicroRNA (miRNA) are important regulators of gene expression and its amendments are related to the development and progression of cancer. Studies of literature and previous results obtained by our laboratory imply some miRNAs in the progression and biochemical recurrence of prostate cancer. The purpose of this study is to research the role of miRNA in the treatment of metastatic prostate cancer models in vitro and in vivo. Method: The lines PC3, DU145 and LNCaP will be used in in vitro experiments. The miRNA and its antiMiRs will be transfected using the lipofectamine. The rate of proliferation and apoptosis will be evaluate by flow cytometry ploidy. Changes in cellular invasiveness will be analyzed using matrigel. Expression levels of target genes RAS, MYC, MMP9, and CD44 SMARCA5 will be evaluated by the technique of quantitative PCR in real time. Athymic mice, males, 5 or 6 weeks will receive intraventricularly injected with strain PC-3M-Luc-C6. On the 20th will be assessed the development of bone metastases with imaging system IVIS in vivo. The animals will be treated with the miRNA and its antiMiR in 23 and 26 and the lesions will be assessed by measuring with the same equipment on days 30, 34, 38, 42 and 44. On day 48 after the last imaging test animals will be sacrificed and their organs analyzed macroscopically and microscopically. (AU)
