Objective: this project aims to observe the expression and distribution of cells of the immune response in gingival tissue from healthy and periodontal disease affected patients through immunohistochemical analysis. Methods: According to the inclusion and exclusion criteria, will be selected 30 patients (10 per group) diagnosed with chronic periodontitis (Group A); aggressive periodontitis (Group B) and clinically healthy gingiva (Group C - control), from which a sample of gingival tissue will be collected during usual dental procedures. Immature dendritic cells will be identified using the markers CD1a, CD207, S100, and mature using markers CD83 and CD 208. Cytotoxic lymphocytes in the epithelium or chorion will be identified using the markers CD3, CD8, granzyme B and perforin. The immunoexpression will be assessed using image representation by a computer system, comprising a light microscope (Leica DM500) adapted to a high resolution camera (Leica ICC50) and a color video monitor. The images will be obtained using the Leica Image Managem IM50 and processed through the program Qwin Leica Image Processing and Analysis System. Independent counts of 10 separate fields for each case will be done. Statistical analysis will be performed using a statistical package previously defined, as well as statistical tests appropriately selected, taking a significance level of p <0.05. The T test will be considered when the results show normal distribution, while the Mann-Whitney test will be considered for the results do not show such a distribution. (AU)
