Abstract

The Mycobacterium chelonae-Mycobacterium abscessus group of rapidly-growing mycobacteria was recently revised, which resulted in the inclusion of new members (M. salmoniphilum e "M. franklinii"), and the reunion of three species species (M. abscessus, M. massiliense and M. bolletii) within a single species - M. abscessus, with the description of two M. abscessus subspecies (subsp. abscessus and subsp. bolletii). Our laboratory was responsible for some of these taxonomic updates. Now, we are studying a group of M. chelonae-M. abscessus isolates without a conclusive species identification. A subgroup includes two isolates from an outbreak in refractive surgery (LASIK) in São Paulo (1999) and one lymph node isolate from a patient from Ribeirão Preto (2007). These isolates showed the same PRA-hsp65 pattern of M. abscessus 1 (BstEII: 210, 235 bp; HaeIII: 145, 70, 60, 55 bp), except for the absence of a 70 bp HaeIII band. Sequence analysis of conserved genes revealed that these isolates were closely related to two zebrafish isolates from USA (JAN1 and JAN2), both without conclusive species identification. Phenotypic and molecular tests using these isolates will be carried out by a PhD student. The present project refers to the genome sequencing and annotation of one isolate (EPM 10906) from this particular subgroup. The objective is to complement the characterization of these isolates and to describe a new species from the M. chelonae-M. abscessus group.
