Abstract

The meningococcal vaccines have been shown effective against serogroups A and C, but still there is no vaccine against serogroup B due to the similarities between the structure and the B capsular polysaccharide acid polisiálico part of the human brain tissue, can lead to autoimmune diseases. The objective of this study is to investigate the properties of a new preparation with outer membrane vesicles (OMPs) of Neisseria meningitidis B, comparing routes of immunization or intramuscular prime-boost in outbred adult mice aged 6-8 weeks using cationic lipid as an adjuvant. Thus, this project aims to identify outer membrane antigens of an epidemic strain of N. meningitidis B and evaluate their interaction with cationic lipid-based adjuvant, aimed at producing formulations with immunoadjuvant activity. As methods of study will be carried out immunoassays such as ELISA, Immunoblot, ELISPOT assessment of humoral and cellular immune response. Antigenic characterization of outer membrane antigens will be done by SDS-PAGE. To characterize the interaction of antigens with adjuvants model cationic lipid techniques will be used for dynamic light scattering, circular dichroism, UV-visible spectrophotometry and fluorescence spectrometry.
