Evaluation of bone repair in the bone/implant interface in osteoporotic rats non treated and treated with alendronate: morphologic and immunohitochemical analysis

Abstract

The purpose of this project is to evaluate the process of bone repair in bone/implant interface in rats with induced osteoporosis treated with raloxifene through Histomorphometric and Immunohistochemical analyses. For this, 30 rats (Rattus norvegicus Albinus, Wistar), with an average weight of 200 grams will be divided into three groups, being Group I (SHAM DN) consisting of 10 rats submitted to sham surgery and fed with a balanced diet; Group II (OVX ST) consisting of 10 rats submitted to bilateral ovariectomy surgery, fed with low calcium diet (osteoporotic) and without medication treatment and Group III (OVX RAL) consisting of 10 rats subjected to bilateral ovariectomy surgery, fed with low calcium diet (osteoporotic) treated with Raloxifene. All of the animals will receive two implants, one in each tibial metaphysis. That will be installed in the right tibia implant with usineded surface and the left tibia implant surface modified by double acid attack. Euthanasia of animals will be performed for 14 days (n = 5/group) and 42 days (n = 5/group) after surgical implant installation through anesthetic overdose. For morphological analysis of the obtained slides, will be performed by hematoxylin and eosin coloration (for assessment of tissue cytoarchitecture) and evaluation of immunolocalization for RUNX2 proteins, osteocalcin, and osteopontin, characterizing the osteoplastic phenotype of bone tissue formed during osseointegration. Immunolocalization is evaluated by assigning scores to be tabulated for comparative evaluation of groups and experimental periods. The data will be submitted to the normal curve to determine the most appropriate test (parametric versus nonparametric). The significance level of 0.05 is used for all tests. (AU)
