Abstract

The characterization of Leptospira strains is essential for understanding the epidemiology of the disease in humans and animals. Traditionally, it is used to characterize the serological classification of the isolates into serogroups and serotypes. However, only the characterization of the genomic of Leptospira species will possible to differentiate them and to understand the molecular mechanisms of the pathogenesis of leptospirosis. The aim of this study was to characterize, by pulsed field gel electrophoresis (PFGE) and comparative analysis of genomes, 36 Leptospira strains isolated from different animal species in different regions of Brazil, in the period 1967-2013. These isolates were previously typed by microscopic agglutination with monoclonal antibodies. However, it was not possible the characterization of the species and serotype, such that are considered potentially autochthonous strains of Leptospira. By PFGE will assess the clonality of strains and genotypic profile of these will be compared with that of known serovars. The isolates with distinct genotypic profile will be submitted to the next-generation sequencing to obtain the respective genomes and it will be compared with Leptospira previously published in GenBank database. Will also be assessed the profile of antimicrobial susceptibility of isolates from the microdilution broth. The results will allow, besides the complete characterization of this collection, species differentiation and new serovars and contribute to the study of the pathogenesis of leptospirosis.
