Antineoplastic potential of NK for onco-hematology treatment in patients undergoing allogeneic hematopoietic stem cells, HLA-compatible and without T-cell depletion

Abstract

Recent results obtained from the genotyping of 50 patients who received hematopoietic stem cell transplantation (HSCT) for onco-hematological treatment from related donors, HLA compatible, without depletion of T lymphocytes and assisted in the Center of Hematology University of Campinas, indicate an increase in overall survival of patients in whom we observed the presence of HLA class I ligands for KIR receptor inhibitor (killer immunoglobulin like) of Natural Killer cells (NK) cells present in the donor and absence to Graft versus Host Disease (GVHD). In this project, it is proposed to evaluate the effect of these NK cells in HSCT, adding determining the haplotype KIR / HLA class I-pair donor patient, the use flow cytometry to determine multicolored to the functional integrity of NK cells and formation of the repertoire of inhibitory receptors (KIR, NKG2A and CD94) along the reconstitution of the immune system after transplantation. This procedure allows to evaluate the algorithm based on the absence of HLA class I ligand in the patient, specific for KIR receptor present on NK cells from the donor to predict whether or not alloreactive NK cells in the graft and also determine the functional integrity these cells prerequisite for promotion of graft versus leukemia effect (EvL)
