Abstract

Dengue is an infectious viral disease transmitted by mosquitoes of the genus Aedes (Aedes aegypti and Aedes abopictus) which has became a major public health problem worldwide. Currently considered a major arboviruses because of its spread to several tropical and subtropical regions where the vector fits best. Approximately 40% of world's population live in endemic areas and the eradication is directly related to the control of its vector. Dengue virus belongs to the Flavivirus genus (Flaviviridae family) and has four antigenically distinct serotypes (DENV-1, DENV-2, DENV-3 and DENV-4) subdivided into genotypes and strains with different degrees of virulence. Several factors influence the spread or the deletion of a genotype and/or strain on population. This project will evaluate the biological characteristics in vitro (replication) and in vivo (virulence) of representatives of DENV-1 genotypes and/or clades, chosen based on previous phylogenetic analyzes. This project will evaluate the biological characteristics in vitro (replication) and in vivo (virulence) of isolates of DENV-1 of São José do Rio Preto-SP, circulating in the years 2008 to 2012, chosen based on previous phylogenetic analyzes. This study will be developed at Laboratório de Pesquisa em Virologia (LPV), FAMERP, with samples of serum from patients with typical symptoms of Dengue that were treated at the hospitals and health care settings in São José do Rio Preto, which is endemic to Dengue. Thus, this study will assist the monitoring of genotypes/strains circulating in the country as well as perform comparisons relating the isolates with virulence degrees and correlates the probable epidemiological consequences.
