Abstract

The aim of BEPE study is to accomplish a comparative acute phase protein (APP) and proteomic studies in milk whey of buffaloes and cows heathy and with mastitis (Study 1), and in blood serum of buffalo calves infected with S. Dublin and S. Typhimurium (Study 2). Study 1: As part of the post-doctoral project accomplished in Brazil (FAPESP Process 2012/10094-0), 252 milk samples from buffaloes and cows were collected and submitted to black sieve cloth test, CMT, SCC and microbiological evaluation. For the realization of BEPE project, the 252 samples will be devided into six groups: milk samples from healthy buffaloes (G1) (n=50), healthy cows (G2) (n=50), buffaloes with subclinical mastitis (G3) (n=50), cows with subclinical mastitis (G4) (n=50), buffaloes with clinical mastitis (G5) (n=2) and cows with clinical mastitis (G6) (n=50). Study 2: As part of PhD project (FAPESP Process 2008/50388-7) and master's degree project (FAPESP Process: 2009/12056-5) accomplished in Brazil, samples from 18 buffalo calves were collected in eight moments, M0 to M8 (before inoculation and every 24 hours after inoculation), being that all animals were also submitted to physical examination and milk samples to microbiological evaluation. For the realization of BEPE project the 18 calves will be divided into three groups: control group (G1) (n=6) that orally received BHI broth, and S. Dublin (G2) (n=6) and S. Typhimurium (G3) (n=6) infected groups, that orally received 108 CFU and 109 CFU of bacteria, respectively, suspended in BHI broth. Study 1 and Study 2: In BEPE project, ELISA analyses of haptoglobin and serum amiloid A (SAA) and proteomic analyses using gel (2D-DiGE: two-dimensional fluorescence difference gel electrophoresis; MALDI-TOF-TOF/MS: matrix assisted laser desorption ionization tandem time-of-flight/mass spectrometry; LC/ESI-MS/MS: liquid chromatography coupled with electrospray ionization tandem mass spectrometry) and gel-free analyses (uHPLC/ESI-MS/MS: Ultra-high performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry) will be performed in the "University of Glasgow".
