Our previous study (Fragoso et al., Food Chem Toxicol., 2013) showed that the intake of acai pulp slowed the development of colon cancer in rats. Therefore, the objective of this project is to study the potential protective mechanisms of açaí (Euterpe oleracea Mart.) In chemical colon carcinogenesis in rats. The animals will be divided into five groups: G1 to G4 will start with two injections of 1,2-dimethylhydrazine (DMH) for two weeks and G5 receive similar injections of EDTA (vehicle DMH). One week after the end of the initiation, the animals receive water plus 2% Dextran Sulfate Sodium (DSS) to induce colitis. After the initial administrations of DMH / DSS, the animals will receive the experimental and control diets until the end of the twenty-fifth week: G1 and G5, basal diet; G2 and G3, basal diet plus 5% and 7.5% lyophilized açaí, G4, basal diet plus 0.2% N-acetylcysteine (NAC) (positive control). The animals will be sacrificed on the twenty-fourth week for evaluation of lesions (ACF and tumors) in the colon. Tumors obtained at the end of the experiment will be used to study gene expression and proteic, and histopathological analysis and study of tumor markers by immunohistochemistry. The freeze dried acai pulp will be characterized for its the phenolic content and total anthocyanin, antioxidant capacity and polyphenolic fractions as present in it. It will also be performed in vitro genotoxicity of fecal water of the animals in Caco-2 tumor cells by the comet assay and the potential for reduced matrigel invasion of Caco-2 cells by cyanidin-3-O-glucoside, the main anthocyanin present in acai pulp.
