The plant Qualea grandiflora Mart., belonging to Vochysiaceae family, is widely found in the Brazilian cerrado, considered a symbol of this region, is used by locals as an anti-inflammatory agent. However, for proper use of medicinal plants is vital knowledge of their toxic and therapeutic potential. Thus the purpose of this study is to evaluate the influence of alcoholic extract of the leaves of Qualea grandiflora Mart. on fibroblasts. For expansion, cells (NIH3T3 fibroblasts) are trypsinized with EDTA (1 mM) and trypsin (0.25%) for 5 minutes at 37°C, followed by inactivation of trypsin with medium containing fetal bovine serum (FBS) . For the experiments are plated 3x10³ cells/well in 96 well plates. After incubation for 24 hours, the culture medium is replaced with alfa-MEM medium supplemented with 10% fetal calf serum (FCS) and different concentrations of the hydroalcoholic crude extract of Qualea grandiflora (pure extract, 1:10, 1:100, 1:1000, 1:10000). After periods of 24, 48, 72 and 96 hours after treatment with different concentrations of the extract isolated fractions, cell viability is measured via the mitochondrial cell activity tests being carried out by the reduction of MTT ( (bromide method 3-(4,5-dimethylthiazol-2-yl) -2,5 - . diphenyltetrazolium) and incorporation of crystal violet values will be analyzed by the Kruskal-Wallis test complemented by ANOVA complemented by Tukey test and Dunn, when obeying the normal or non- normal distribution, respectively. significance level adopted will be 5% (p<0.05).
