Abstract

Aging related diseases are the leading cause of death in the world. This study aims to investigate molecular aspects of innate immunity in the senescence process, and as a predisposition factor for the development of sepsis, a complex disease associated with aging. We will perform a broad gene expression analysis to identify genes with a critical role in the aging process and to investigate differences in immune and inflammatory responses between young and old individuals. We will evaluate the expression of protein-coding mRNAs, long noncoding RNAs (lncRNAs, >200 nt), yet unreported in the literature, and microRNAs. To generate gene expression profiles, RNA isolated from neutrophils collected from blood samples from young and old subjects, with and without sepsis, will be hybridized with high-density DNA microarrays. The data extracted from microarrays will be computationally analyzed to select genes, lncRNAs, microRNAs, and molecular pathways differentially expressed during senescence and sepsis. After extensive functional annotation, data will be stored in a relational database to facilitate data mining of candidates for detailed characterization in the laboratory. (AU)
