Abstract Plants displayed a fundamental importance in the lives of all people. Natural products extracted from plants, have emerged as important sources of new therapeutic agents used in the treatment of several diseases, including cancer. Therefore, several studies were conducted to analysis of active principles of plants, with selective tumor cell activity, as well as investigating the mechanism of action of drugs with pharmacological action. Thus, this project aims to identify, isolate and elucidate the structures of compounds which showed cytotoxic activity from ethanol extract of leaves of Guarea macrophylla ssp. tuberculata (Meliaceae) against several cell lines (melanoma, breast adenocarcinoma, leukemia and human carcinoma). To conduct this study, the bioassay-guided phytochemical fractionation will be employed to the isolation and structural characterization of the active compounds present in the extracts. Furthermore, we intend to elucidate the mechanism of action related to tumor cell apoptosis through tests performed in vitro by evaluating the degradation of DNA, tunel assay, analysis of apoptotic morphological changes by transmission electron microscopy - (TEM), determination of mitochondrial membrane potential (Èm) and determination of intracellular superoxide anion.
