Gene expression of IL6 and TNFA in individuals with type 2 Diabetes Mellitus, Dyslipidemia and Periodontal Disease

Abstract

Diabetes mellitus type 2 (DM2) and Dyslipidemia systemic diseases are involved in the pathogenesis of periodontal disease (PD) and may alter the immune-inflammatory response of the host. The objective of this study is to investigate the expression of proinflammatory genes interleukin 6 (IL6) and Tumor Necrosis Factor Alpha (TNFA) in patients with DM2 (compensated and decompensated), Dyslipidemia and DP. Yet wasn't seen in the literature any study investigating the expression of these genes in patients who manifest simultaneously the three diseases mentioned. Prior to this research project, our team investigated five groups of patients (30 patients each): DMdDisDP (decompensated type 2 diabetes, dyslipidemia and periodontal disease), DMcDisDP (compensated diabetes, dyslipidemia and periodontal disease), DisDP (only dyslipidemia and periodontal disease), cDP (only periodontal disease) and control (without any of the three diseases). All patients underwent a complete periodontal examination, physical examination and biochemical evaluation of glycemic and lipid profiles. From each patient, blood was collected, and the RNA extracted. In the current design is made of all cDNA samples for investigation of the expression of IL6 and TNFA gene by Real-Time PCR (qPCR) using the SYBR Green system. It is intended to better understand the pattern of expression of these important pro-inflammatory genes, and subsequently confront these results with our previous analysis of the expression of anti-inflammatory IL10 gene. This will allow us to understand the possible interrelationship of these genes and their influence on glucose, lipid profile and periodontal patients with the aforementioned conditions.
