Abstract

Neonatal anoxia is one of the most commom causes of morbidity and mortality in neonates, especially in preterm infants, constituting an important public heath problem due to permanent sequelae it presents. The oxygen deprivation triggers series of cascades simultaneously, culminating in cell death, especially in metabolically vulnerable regions such as the hippocampus. In cell death by deprivation of oxygen process, calcium plays a crucial role, since its massive entry within cell leads to activation of glutamatergic receptors, culminating in death by excitotoxicity. In this study, we investigated the role of the intracellular calcium receptors inositol 1,4,5-triphosphate through analysis of its gene expression, semiquantitative analysis of IP3R-postive cells and histological (apoptosis) and behavioral (spatial memory and learning) consequences of the use of blockers such as 2-APB, reversible inhibitor of opening IP3R permeable to cells, at three timepoints after anoxia: 6 and 24 hours for histological and molecular analyses and P60 for behavioral testing. We expected, with this project, provide basis for the development of therapeutic proposals that minimizes the injuries caused by oxygen deprivation and its functional sequelaes. (AU)
