Abstract

Cryopreservation of fish gametes is an important tool in the conservation of genetic material from species, especially endangered ones like the surubim-do-Paraíba, Steindachneridion parahybae. In the present study will be evaluated aspects to cryopreservation of oocytes, aiming to make a satisfactory protocol. The toxicities of oocytes at different concentrations of cryoprotectants tests will be conducted: Methanol, ethylene glycol and sucrose. Early stages, vitellogenic and mature oocytes of the surubim-do-Paraíba be hatched in different maintenance medium (Hank and L-15). The cryoprotectant solutions (CPAs) will be prepared in the same medium. After toxicity testing, the CPA will be defined and the oocytes undergo this solution. Then pass through the cooling curve and stored for 30 and 60 minutes at -5°. From the results obtained, the stage of the oocyte to be used will be chosen. Therefore, will be tested in different concentrations of CPA storage oocytes in liquid nitrogen (-196 ° C) for at least 24 hours. Tests for viability of oocytes are: analysis of membrane integrity using a vital dye (Trypan Blue); maturation in vitro by observation of germinal vesicle breakdown (GVBD); and fertilization of mature oocytes.
