Abstract

We will explore the hypothesis that sleep loss induces cellular damage, leading to up-regulation of inflammatory markers, down-regulation of lipid transport pathways and up-regulation of fatty acid and triglyceride synthesis, favoring cardiometabolic diseases. Pharmacological approach will be used to investigate the role of the NLRP3 and LXR activation and inactivation on the inflammatory and lipid metabolism transport pathways as a function of sleep-wake behavioral states and after sleep deprivation. Rats will be sleep deprived using the gentle handling method. Agonists or antagonists will be administrated before and during sleep deprivation. At the end of deprivation, blood and tissue samples (brain and liver) will be collected for analysis. The tissues will be analyzed for gene expression of lipid metabolism and transport pathways as well as inflammation-related genes. During the experiment, the spontaneous sleep-wake cycle as well as sleep deprivation and sleep opportunity after the deprivation will be monitored using EEG/EMG recording. Understanding the mechanisms by which sleep deprivation induces activation of the immune responses and metabolic changes could help us to understand the relation between sleep loss and adverse health effects.
