The effects of low-level laser therapy at different doses in bone repair of critical size defects: histomorphometric study in rats

Abstract

This study aims to investigate the effects of low-level laser therapy (LLLT) in different doses, applied in bone defects of critical size by histological and histomorphometric analysis. Sixty adult rats (Rattus norvegicus, Albinus variation, Wistar) will be used, with 90 days of age, weighing approximately 300g. Critical defects will be made in the skull with 5 mm of diameter. The animals will be randomly divided into 5 groups: G1-bone defect + blood clot, G2-bone defect +LLLT 15 J / cm2, G3- bone defect + LLLT 30 J / cm2, G4-bone defect + LLLT 45 J /cm2, G5-bone defect + LLLT 60 J / cm2. Each group will be divided in accordance with the periods of observation, 30 and 60 days, with 6 animals in each subgroup. A low power GaAlAs with wavelength of 660 nm, 30 mW area of 0.04 cm2 will be used. LLLT is applied on 5 points so that any surgical wound uniformly receives treatment. Four application points are distributed along the edges of the wound in clockwise 3, 6, 9 and 12 hours, respectively. And a point of application located in the central region of the surgical wound. The laser irradiation will take place immediately after the procedure. On 30 and 60 days, six animals from each group will be sacrificed by anesthesia overdose. Then the calvarias will be removed for histological and histomorphometric analyses. All data will be statistically analyzed by ANOVA complemented by Tukey test to verify the statistical differences between groups. The level of significance will be 5%. (AU)
