Abstract

Frailty is a condition that often results in the decline of physiological systems and cause vulnerability to adverse conditions because of difficulties to keep homeostasis in situations of exposure to disturbances. The syndrome has been recognized as a significant public health and clinical challenge. Molecular biomarkers directly or indirectly related to the pathophysiology of multiple diseases related to frailty context have been identified. Among them, studies showed changes in immune profiles, as well as deregulation of microRNAs (miRNAs)-related gene expression of molecules involved in the pro-inflammatory activity. This project is a cross-sectional study, based on the assumptions of quantitative research, which aims to determine whether specific markers of chronic low-grade inflammation, as well as for gene expression of miRNA in blood samples are altered in frail elderly subjects. The research will be developed in São Carlos, headquarters of the Federal University of São Carlos (UFSCar). Participants will be invited to participate in the study, older adults living in areas of higher social vulnerability of São Carlos, SP that are on the roof of the Support Center for Family Health (NASF). Biological samples for research subjects will be collected, analyzed and subsequently stored in a biorepository. The whole blood of subjects will be studied by qPCR techniques, by differential expression compared with non-fragile subject. Peripheral concentrations of biomarkers will be determined by enzyme-linked immunosorbent assay (ELISA). Statistical tests of association, regression and diagnostic accuracy will be realized. The results may contribute to the deployment of tools that work in the diagnosis/prevention of frailty syndrome.
