Abstract

The search to identify new drugs in toxins produced by animals, like snake, spider and bee venoms, has increased greatly in recent years. In this context, different approaches are used to obtain bioaffinity ligand screening with low time/sample-consuming and accurateresults, by affinity and activity assays.The aim of this work is the screening of peptides, in toxins, as acetylcholinesterase ligands. Acetylcholinesterase was the selected target and the Electrophorus eletricus the species nominated as model. The enzyme will be immobilized onto the surface of amine terminated magnetic beads, producing bioreactors. Bioaffinity screening assays using the ligand-fishing approach will be adopt to identify potential peptides as ligands and, the compounds fished-out will be sequence and identified by nanoLC-MS/MS. For orthogonal validation, the experiments will be carried out with the reference peptides ligands: fasciculins I and II, extracted of green mamba snake venom.
