BIOSTIMULATION OF PULP CELLS BY ADDITION OF SIMVASTATIN IN GLASS IONOMER CEMENTS

Abstract

The stimulation of the dental pulp regenerative process by modulating cell responses is a very important characteristic of dental materials used on deep dentin and exposed pulp treatment. Therefore, the aim of this project is to evaluate the effectiveness of simvastatin incorporation to glass ionomer cements (GICs) on odontoblast-like cells secretive activity stimulation. Initially, a bioactive and cytocompatible concentration of simvastatin will be determined (0.01 µM/L; 0.1 µM/L; 1 µM/L), by mensurating the cell viability (Alamar Blue), alkaline phosphatase (ALP) activity (thymolphthalein monophosphate) and mineralized matrix deposition (Alizarin Red) of odontoblast-like MDPC-23 cells. According to the results, the simvastatin will be incorporated to conventional and resin-modified GICs and its release in humid environment will be assessed (absorbance 428 nm). Then, the bioactivity to the odontoblast-like cells of the GICs containing or not simvastatin will be assessed. For that, the MDPC-23 cells will be seeded on the lower membrane of transwells with 3um poro size (simulating the size of dentin tubules at deep dentin), and the materials will be applied on the upper membrane surface. The cell viability, ALP activity and mineralized nodule deposition will be evaluated at different time-points. The data will be analyzed regarding normality and homoscedasticity in order to select the statistics tests (p = 0.05).
