Abstract

Life is dependent on proteins; each step of generating cell, since replication of the genetic material to senescence and death, depends upon the proper function of several proteins, which demonstrates and justifies the importance of the study of these biomolecules. Proteomics can be defined as the direct qualitative and quantitative analysis of all the proteins in an organism, tissue or cell under a given set of physiological and environmental conditions. Recent advances in mass spectrometers and liquid chromatography is providing numerous approaches in the study of proteomics, including those related to basic and clinical research involving stem cells. In this context, Dr. Eliandre Oliveira Cacheado accumulated great experience in proteomic analysis, being the Head of "Proteomics Platform of the Barcelona Science Park" (Parc Científic de Barcelona, PCB), renowned center in Barcelona, Spain since 2002. In addition, the researcher is also responsible and collaborator in projects of the European Commission (ShockOmics and Heals), besides the publications in renowned journals in the proteomics area, such as "Proteomics and Journal of Proteome Research that have a high impact factor (JCR ). The supervision of the researcher in the internship will be of great learning and improvement to post-doc, because besides the development of the proposed project, he will be able to accompany and participate in the projects of the European Commission (ShockOmics and Heals) conducted by a group with extensive experience in proteomic analysis. The internship will allow the research training in: (1) sample preparation; (2) identification of proteins through enzymatic digestion, analysis with high resolution mass spectrometry (LC-MS/MS) and search in databases using different search engines; (3) identification and relative quantification of the level of protein abundance by LC-MS/MS and (4) bioinformatics tools. The experience with research training will contributes for the improvement of quality and internationalization of our current and future works. In addition, the supervision of Dr Eliandre, the collaboration of her team and the excellent infrastructure of the PCB will contribute directly to the technical improvement of proteomic analysis of equine mesenchymal stem cells from umbilical cord (MSCs-UC) and bone marrow (MSCs-BM) fresh and cryopreserved in the proposed project of post doctoral named "Comparative study of the effect of cryopreservation on the biological properties of equine mesenchymal stem cells from umbilical cord matrix and bone marrow". For this, samples of cell lysates (protein extracts) obtained from equine MSCs-BM and MSCs-UC fresh and cryopreserved with different freezing media will be subjected to reduction, alkylation, tryptic digestion and subsequent analysis LC-MS / MS and date analysis for protein identification and quantification.
