Abstract

Recently, our group developed an ultra-sensitive technology based on HPLC / micro HPLC / mass spectrometry to measure DNA oxidation and modifications induced by reactive aldehydes. Our main project is to investigate the use of these lesions as redox stress biomarkers. The present project aims to compare DNA lesions (8-oxo-7,8-dihydro-2'-deoxyguanosine, 1,N2-ethene-2'-deoxyguanosine and 1,N2 propane-2'-deoxyguanosina in cell model for amyotrophic lateral sclerosis (ALS) and Xeroderma pigmentosum (XP), where redox stress is involved. The level of aldehydes in control and in cells exposed to stress will also be evaluated. Possible effect of molecules that participate in aldehyde detoxification (glutathione and carnosine) will also be investigated. The study with Xeroderma pigmentosum model cells will be carried out in collaboration with Prof. Nadja Cristina de Souza Pinto from our Department.
