Abstract

In recent years, silver microcrystals have become the focus of studies. One of the evidenced properties of these materials is its antimicrobial activity against different microorganisms, due to the presence of silver in its composition. In this study, the antifungal activity of silver alpha vanadate (±-AgVO3) against Candida albicans (ATCC 90028) and its cytotoxicity on spontaneously immortalized normal oral keratinocytes (NOK-si) cells will be investigated. The microcristiffs of ±-AgVO3 will be synthesized by the co-precipitation method under three different temperatures (10, 20 and 30ºC) and characterized by x-ray diffraction, field emission scanning electron microscopy and Raman spectroscopy. The antifungal activity will be evaluated from serial microdilution of microcrystals (according to the Clinical & Laboratory Standards Institute - CLSI), where minimum inhibitory (MIC) and fungicide (MFC) concentrations will be determined. Fluorescence microscopy images with microcrystals at the minimum inhibitory and fungicidal concentrations will be obtained in order to confirm the microbiological findings. The cell viability of NOK-si cells will be evaluated through the Alamar Blue assay, and scanning electron microscopy (SEM) images of all the evaluated groups will be performed. In cell assays only four concentrations of microcrystals (MIC, MFC, MIC diluted 10 times and 10 times concentrated) will be used. The results obtained will be submitted to statistical analysis for further interpretation and discussion.
