FGF2, VEGF, DSPP, and DMP-1 expression in cells of the pulp deciduous teeth with a low intensity laser

The objective of this study is to investigate the Low Intensity Laser effects of different energy densities on viability of the pulp of human deciduous teeth; mRNA expression of FGF2, VEGF, DSPP and DMP-1; and expression of the FGF2, VEGF, DSPP and DMP-1 protein. Human deciduous teeth samples of pulp tissue will be obtained and grown in specific culture. After the fourth passage, the sample will be irradiated with low intensity laser to the following groups: Group I - Ia (1.2 J/cm2 - 05 mW - 10s), Ib (2.5 J/cm2 - 05 mW - 20s), Ic (3.7 J/cm2 - 05 mW - 30s), Id (5.0 J/cm2 - 05 mW - 40s), ie (6.2 J/cm2 - 05 mW - 50s), if (no irradiated); Group II - IIa (1.2 J/cm2 - 05 mW - 10s), IIb (2.5 J/cm2 - 10 mW - 10s), IIc (3.7 J/cm2 - 20 mW - 10s), IId ( 5.0 J/cm2 - 30 mW - 10 sec), IIe (6.2 J/cm2 - 40 mW - 10 sec), IIF (no irradiated). The techniques used for the analysis will be MTT, Crystal Violet, RT-PCR and ELISA. The values in triplicate will be analyzed by ANOVA followed by Tukey test and Kruskal-Wallis analysis complemented by Dunn's test when obey normal and non-normal distribution, respectively. The significance level of 5% will be adopted for the differences considered statistically significant. (AU)
