Abstract

The aim of this project is to synthesize and characterize simvastatin incorporated nanofibers scaffolds, evaluating its properties and biological response. The experiments will happen in in two phases: The first phase will involve the synthesis and characterization (i.e., morphology, mechanical strength, and drug release) of simvastatin incorporated nanofibers scaffolds using two biodegradable polymers of distinct molecular weights (i.e., PLGA50-6.5K, PLGA50-64K). The second phase will be dedicated to evaluate dental pulp stem cell-scaffold interactions. Besides that, will also be evaluated cell proliferation, viability (MTT or XTT), mineralization by Alizarin Red method, cell adhesion/penetration (Confocal microscopy), and odontoblastic differentiation using selected markers, alkaline phosphatase content, osteocalcin, bone sialoprotein, collagen 1, dentin sialophosphoprotein-DSPP. The data will be submitted to descriptive and inferential statistical analysis according its normality with parametric or non-parametric tests. (AU)
