Abstract

Background: Preterm Labor (PTL) and Preterm Premature Rupture of Membranes (PPROM) are responsible for severe perinatal morbidity and neonatal mortality worldwide. MicroRNAs (miRNAs) can affect gestational outcome are potencial biomarkers for these complications. Exosomes act in cell comunication, contain miRNAs and increase their stability. Objective: To compare the expression, in exosomes from peripheral blood, of the miRNAs miR-15-5p, miR-143-3p, miR-145-5p, miR-199a-5p, miR-200c-3p, miR-223-3p, miR338-3p and miR-493-3p between women with term pregnancies and PTL or PPROM, and to elucidate possible mechanisms of action of these miRNAs in the physiopathology of these adverse outcomes. Patients and methods: Case-control study with women that underwent PTL, PPROM, term pregnancy with and without labor and non-pregnant women seen at Botucatu Medical School. Exosomes will be isolated from plasma and expression of miRNAs will be performed by Real Time PCR. Later, miR-mimics or anti-miRs of miRNAs differentially expressed will be used to challenge cultures of chorioamniotic membranes. Pellets and supernatants from the cultures will be used to evaluate gene (pellets only) and protein expression of predicted targets of miRNAs. Levels of the cytokines IL-1², IL-6 and IL-8 will be measured in the supernatants. Statistical analysis will be performed according to the results obtained and a p-value of 5% will be adopted. (AU)
