Abstract

It has been suggested that there are histological and functional distinctions between the periodontal ligament (PDL) of deciduous (DecPDL) and permanent (PermPDL) teeth. Thus, we hypothesized that DecPDL and PermPDL display differences in the constitutive expression of genes/proteins involved with PDL homeostasis. Primary PDL cell cultures will be obtained for DecPDL (n=3) and PermPDL (n=3) to allow us to perform label-free quantitative secretome analysis. Next, total RNA and protein extracts will be obtained from fresh PDL tissues of deciduous (n=6) and permanent (n=6) teeth, and Western blotting and qPCR analysis were used to validate our in vitro findings. In order to systematically acess potential morphological differences between DecPDL and PermPDL, we will use polarizing and light microscopy analysis in histological sections obtained from minipigs. The fold-change calculation will be based on the ratio of the mean of the normalized counts of PermPDL and DecPDL [mean PermPDL / mean DecPDL] and the beta-binomial model for analysis of spectrum counts to determine the differentially and exclusively proteins expressed. Data from protein (Western Blotting) and gene (qPCR) expression were analyzed by Student´s t test (±d0,05), while the histological results will be presented descriptively.
