Abstract

The mechanisms of genetic contribution to development of obesity need to be established in interventions that aim to weight loss, as bariatric surgery. Therefore, the analysis of gene expression by microarray related energy expenditure and subsequent association with the resting metabolic rate and energy metabolism may elucidate the understanding of genetic factors that contribute to obesity. The aim of this study is analyze molecular markers related to energy metabolism and substrate oxidation associated with measures and resting metabolic rate in women with obesity III grade before and after bariatric surgery. The sample consisted of 20 women of mixed population, aged 18 to 60 years, divided into two groups: group 1 (G1) 10 women with obesity III grade candidates for bariatric surgery followed at the Bariatric Surgery Clinic of HCFMRP- USP and group 2 (G2) control group of 10 normal. Nutritional assessment will be made by Body Mass Index (BMI), body composition by bioelectrical impedance method, Resting Metabolic Rate (RMR), glucose oxidation and lipid by calorimetry indirect. To assess genetic profile, will be collected peripheral blood and use microarray methodology for BeadChip and then validate by quantitative PCR method in Real Time. After six month, participants underwent bariatric surgery will be re-evaluated and compared with control group, pre and post surgery. Results will be analyzed by SPSS software version 17.0 and the level of significance will be established at p <0.05.
