Abstract

Human T cell lymphotropic virus type 1 (HTLV-1) was the first human retrovirus to be discovered. Approximately, 10 - 20 million people are infected with HTLV-1 worldwide, and in Brazil, there are about 2.5 million HTLV-1-carriers. The main HTLV-1-associated diseases are the HTLV-1 associated myelopathy/tropical spastic paraparesis (HAM/TSP) and the adult T cell leukemia/lymphoma (ATL). Currently, there is no cure and no preventive vaccine for this infection. Also, the therapeutic approaches for the treatment of HAM/TSP have variable effectiveness. Thus, the identification of new compounds with antiviral action, mainly by high content screening assays (HCS), is necessary. Some studies have adopted the construction of reporter cell lines based on the green fluorescent protein (GFP) gene expression under the control of the viral promoter region, since these tools are fast, easy and inexpensive for antiviral drug studies. Therefore, the aim of this project is to construct a HTLV-1-infected reporter cell line which carries the full viral genome and the GFP gene under the control of the long terminal repeat (LTR) promoter (Jurkat-HTLV-1-LTR-GFP) for use in antiviral screening assays. The construction of this reporter cell line will be a powerful strategy for the identification of new antiviral compounds with inhibitory action on HTLV-1 gene expression.
