Abstract In this study, the biocompatibility and bioactivity with human dental pulp cells of a chitosan-collagen scaffold containing calcium aluminate mineral phase will be evaluated. The scaffold will be prepared by mixing a chitosan solution (2%) with collagen gel (4 mg/mL) in a 2:1 ratio (v/v), followed by phase separation at -80oC and freeze-drying. A chitosan-collagen scaffold with no calcium aluminate will be prepared. Monolayer cells seeded onto inert material (polystyrene) will be used as negative control. First of all, the active cell migration mediated by the materials will be assessed. For that, the human dental pulp cells will be seeded on 8 um pore size membrane of trans-well devices, and this set will be positioned in intimate contact with the materials. Cell migration will be assessed after 4- and 24-hour incubation time by light microscopy and scanning electron microscopy. Cell viability will be monitored after 1, 7 and 28 days of cell seeding onto material surface, by means of live/dead assay. Finally, micro-CT analysis will be performed on materials before and 28 days after cell culture in order to determinate the mineral contend of scaffolds. Numerical data will be subjected to specific statistical analysis. | |
