Head and neck cancer is the sixth most common cancer worldwide, and oral cavity it the most frequent, followed by larynx and pharynx. The presence of microscopic tumor in surgical margins contribute to increased local recurrence rate and decreased overall survival. From 15 to 20% of patients with negative histological margins will present locoregional recurrences. In these cases, surgical resection may not have been able to remove all the tumor cells. Many studies have suggested the presence of genetic and molecular changes prior to the onset cancer phenotype in the mucosa adjacent to the primary tumor. Aberrant DNA methylation patterns are found in many diseases, especially in cancer, where there is a loss of DNA methylation control. Increased methylation in the promoter region of a tumor suppressor gene can lead to progressive reduction of its expression, resulting in silencing and selection of proliferative advantage cells. Therefore, this project aims to determine the hypermethylation profile of genes in oral cavity squamous cell carcinoma samples (OSCC) and its surgical margins with negative histology. For this, there will be an initial screening by Illumina equipment in 15 paired samples (tumor and 2 margins) for the selection of candidate genes, and subsequent validation by an additional 35 paired samples in the same way, by Pyromark Q96ID equipment. For statistical analysis will be used chi-square test or Student t test, Fisher's exact test and Kaplan-Meier by SPSS 21.0 software. (AU)
