Abstract

Pulp revascularization has resulted in more favorable prognosis compared to apicification at the treatment of immature necrotic teeth. Remaining cells from apical papilla have been suggested as the key for the treatment success. However, the molecular mechanisms involved with the modulation of these cells under unfavorable environment such as inflammation remains unclear. Among the mediators involved in this process, angiotensin II (AngII) might be suggested as an important modulator of cellular function. The aim of this study is to elucidate the role of AngII at the osteo/odontogenic differentiation of human apical papilla cells (CPA) in vitro. CPA cultures will be established from third molars with incomplete root formation and characterized. The cells will be subjected to stimulum with increasing concentrations of lipopolysaccharide (LPS) for cellular viability assay. Then, CPA will be challenged with LPS at proliferation (MP) or differentiation medium (MD) for detection of Ang II production and the receptor AT1. Its role at cellular differentiation will be investigated by means of pharmacological blockade. Data will be analyzed by means of GraphPad Prism 5.0 using One-way analysis of variance (ANOVA) followed by Tukey´s post-test. Significant values will be set at p < 0.05. (AU)
