Effect of different commercial mouth rinses containing alcohol on the viability of microcosm biofilm and on enamel demineralization prevention

Abstract

Different antimicrobial agents with alcohol or alcohol-free are used to control dental biofilm, among them cetylpyridinium chloride, essential oils, xylitol, fluoride, triclosan and chlorhexidine. These commercial mouthrinses are commonly used by population, without prescription, with the aim of controlling halitosis. The active agents included into commercial mouthrinses can modify the metabolism of microorganisms inhibiting the production of acids that cause tooth demineralization; however, there are few comparative studies of the different active agents. This study will evaluate the effect of these commercial mouthrinses on the viability of a microcosm biofilm as well as on the prevention of enamel demineralization. For the formation of a microcosm biofilm, saliva will be collected from 2 healthy subjects, who have not brushed their teeth in the last 24 hours and have not ingested food or drinking liquids in the last 2 hours. Saliva will be diluted (70% saliva and 30% glycerol) and aliquots of 1 ml of saliva will be stored at -80° C and further mixture to McBain saliva (with 0.2% sucrose) for the formation of a cariogenic biofilm. Bovine enamel samples (4 mm x 4 mm) will be prepared for the formation of a microcosm biofilm for 7 or 14 days, which will be treated daily (1x60s/day) using the following commercial mouthrinses: PerioGard®, Noplak®, Listerine®, Malvatricin® and Cepacol®. In Step 1, live and dead bacteria in the biofilm will be evidenced by fluorescence using confocal microscope. In Step 2, the demineralization caused by the biofilm in the studied conditions will be measured by transverse microradiography. The data will be subjected to appropriate statistical analysis (parametric or non-parametric test, p <0.05). (AU)
