Abstract

Rheumatoid arthritis (RA) is considered an autoimmune disease, characterized by chronic inflammation in the hyperplastic synovial membrane and bone tissue, causing severe joint loss, which is predominantly generated by the increase in the number of osteoclasts. Leukotrienes (LTs) are lipid mediators derived from arachidonic acid via the 5-lipoxygenase (5-Lo) pathway and play an important role in inflammatory response as well as bone resorption. In addition, LTs are synthesized by osteoblasts and osteoclasts. However, although they are involved with the inflammatory process in arthritis and little is known about an individual's contribution, the functional and molecular parameters, the cells to mediate, through LTs, a bone resorption in that condition. (CIA) in 129 / Sv WT mice (wild-type) and 5-Lo KO (knockout to 5-Lo), and to identify the profile of the colors involved in the art as well as the histomorphometric parameters of the tissues involved in this condition. For this, the mice are immunized intradermally with 100 ¼l of emulsion (100 ¼g collagen) and after 28-40 days of immunization, the edema will be quantified by measuring the thickness of the hind paw with a calibrator. Four groups will be analyzed: control and treated (129 / Sv WT and 5-LO KO induced or not by collagen). Bone marrow specimens (cartilage, vascular endothelium and bones) were collected for histological assays, densitometry analysis (computerized micro-tomography), immunohistochemistry to evaluate the distribution of studies of interest and tissue analysis for proteomic analysis. For a validation of the proteome results the Western blotting of the most expressed targets must be performed. Therefore, this study should contribute to a better understanding of the role of LTs involved in the mechanisms of rheumatoid arthritis.
