Abstract

The project focuses on the manufacturing of polymeric vesicles (polymersomes) for gene and protein encapsulation and delivery. The first step aims the production and structural characterization of assemblies produced from the block copolymers PEO45-b-PCL118, PEO45-b-PLA174, PHPMAm-b-PDPAn and PEOm-b-PDPAn. The synthesis of the PDPA-based block copolymers is intended to be performed at the Centre of BioMedical Polymers (Czech Republic) during a BEPE internship. The structural characterization will be performed essentially via scattering techniques (DLS, SLS, ELS, SAXS) and imaging (TEM). Subsequently, the encapsulation and release of biomacromolecules (proteins and nucleic acid) will be investigated and further, interaction of the polymeric vesicles with biological system (cells) will be evaluated via MTT cytotoxicity, cellular uptake and intracellular release experiments. Similar investigations will be performed by using the block copolymer functionalized with the peptide sequence RGD (RGD-PHPMAm-b-PDPAn). (AU)
