Abstract

The species from family Cactaceae are associated to arid and semi-arid climates, becoming attractive models to study evolutionary process in xeric environments. The main goal of this project is isolate molecular data from nuclear and plastid genomes, which might be useful to infer phylogenetic relationships of genus Cereus (Cactaceae) species. Thus, it will be performed the cross-amplification of primers set previously described to amplify plastid segments and know genes as well as some anonymous nuclear loci recently described for the cactus genus Pilosocereus. To get molecular information an amplicons library will be prepared according protocols available in literature, followed by massive DNA sequencing in MiSeq Illumina platform. The generate data will be analyzed following traditional phylogenetic (Maximum Likelihood and Bayesian Inference) and coalescent-based analyses (Species Tree). Overall, this project should contribute for both the standardization of the method to prepare amplicons library as well as to elucidate phylogenetic relationships of genus Cereus. (AU)
