Associated to the scholarship: | 16/03245-2 - Peri-implant bone healing in the tibia of spontaneously hypertensive rats (SHR) treated with losartan: a biomechanical, molecular, microtomography, bone dynamics by fluorochromes, immunohistochemical and histological analysis, BP.MS |
Abstract The aim of this study is to evaluate the peri-implant bone healing process by histomorphometric analysis and investigate the influences that losartan treatment may lead to peri-implant bone healing. The influence of losartan in the peri-implant bone healing will be investigated in 32 tibias of male rats; tibias was collected from 16 spontaneously hypertensive rats (SHR) and 48 normotensives (Wistar) that received one implant in the tibia. The animals were divided into groups treated and not treated with losartan (30 mg/kg/day). With 14 and 42 days, they received the application of fluorochromes (calcein/alizarin red), respectively. The animals were euthanasia after 60 days of implant placement and their tibias were collected. The tibias will be submitted to inclusion process in light-curing resin for making the slides in the calcified cutting system EXAKT; The sections will be subjected to staining Levai-Laczko stain, and scanned to measure the percentage of newly formed bone tissue per tissue volume (nBV/TV), the percentage of old cortical bone per tissue volume (oBV/TV) and the bone to implant-contact (BIC) in the region of interest (ROI), and then, the same samples will be used by epifluorescene analysis, to analyse the bone dynamics at 14 and 42 days marked by the application of fluorochromes (calcein/alizarin red); for this, the images taken by microscope will be submitted to the measurement of fluorochromes precipitation areas (Calcein and Alizarin) in the peri-implant region. All quantitative results will be statistically analyzed adopting a 5% significance level. (AU) | |
