Effects of mechanical vibration on IL-1b and TNF-a levels in periodontal ligament cells

Abstract

Mechanical vibration has been shown to be efficient in bone and tissue repair, because through mechanical stimuli, cellular alterations occur, which can stimulate or inhibit cells. In addition, the vibration has been used in orthodontic treatment, and may influence the tooth movement. There are devices on the market that emit vibrations for use during orthodontic treatment. AcceleDent (OrthoAccel® Technologies, Houston, Texas, USA) produces 30 Hz and should be used daily for 20 minutes. This device is already in the market, but to the best of our knowledge, there are no studies in the literature about the effects of mechanical vibration in periodontal disease. The objective of this work is to evaluate the mechanical vibration protocol used by AcceleDent in Porphyromonas gingivalis LPS-challenged periodontal ligament cells, simulating periodontal disease in vitro. LPS-challenged periodontal ligament fibroblast cells (hPdLF) will be cultured. After the addition of LPS (or PBS), the plates will receive the vibration protocol (30 Hz for 20 min), and 2 hours after the beginning of the vibration, the cell viability will be evaluated and the supernatant will be collected later to quantify IL-1² and TNF-±. The same vibration procedure will be repeated 24 h and 48 h after the addition of LPS / PBS, and the analyzes will occur 2 h after the first and third vibration sessions. The data will be submitted to the most convenient statistical test, at the 5% level. (AU)
