Abstract

Ovarian cancer is the fifth death cause regarding malignant neoplasms in women. Thereby, in recent years, efforts have been made to demonstrate that inherited genetic markers in women with ovarian carcinoma, such as polymorphisms, may predict the response, resistance and toxicity to chemotherapy with platinum derivatives and paclitaxel, and survival. Furthermore, in ovarian carcinoma, surgery and chemotherapy can affect the quality of life. This project aims to evaluate 1) the association between inherited genetic markers in women with ovarian carcinoma with chemotherapy response and adverse reactions with carboplatin and paclitaxel, progression-free survival, overall survival; 2) and new cases evaluate the relationship between variables already cited and quality of life. This project is an observational cohort study. The study will include women, who will receive carboplatin and paclitaxel as treatment of disease, in the Women's Hospital (CAISM) at State of University of Campinas (UNICAMP). For the purpose of 1) 136 women with ovarian carcinoma and her biological material will be included by signing the Informed Consent and Informed of the Biobank of Comprehensive Care Center for Women's Health (CAISM-UNICAMP) (CONEP B -056). Genetic markers corresponding to detoxification, DNA repair, apoptosis related cell and angiogenesis polymorphisms, will be analyzed by PCR and enzymatic digestion or real time PCR in DNA extracted from peripheral blood. All adverse reactions and toxicities detected will be documented and classified according to their severity. For the purpose 2) will be applied a questionnaire of quality of life in women with ovarian carcinoma attended as new cases and begin chemotherapy with carboplatin and paclitaxel. The questionnaire will be administered before chemotherapy, in the fourth and sixth cycle. Quality of life questionnaire will be applied in women with ovarian carcinoma classified as new cases in the begin of treatment with carboplatin and paclitaxel. Questionnaire application will be repeated in the fourth, sixth cycle of chemotherapy and one year after the end of chemotherapy. Demographic and clinical data of the study participants will be obtained from the medical records. The data acquired and recorded in the collection form will be transferred to Microsoft Excel spreadsheet. The data will be analyzed using the statistical package R. The significance level will be 5%.
