Abstract

The increase of microbial resistance to known antimicrobials and the difficulty to eradicate microorganism growing in biofilms indicate a need for searching novel natural agents with antimicrobial and antibiofilm activities. A rich resource of these agents are plant extracts and fractions. In the oral cavity, biofilm formation initiates with planktonic microorganisms adhering to the salivary pellicle on teeth and with the formation of the initial extracellular matrix, which is primarily composed of glucans synthetized by glucosiltransferases. Therefore, the aim of this study is to determine the antimicrobial and antibiofilm effect of extracts and fractions from plant species (including different accessions of Casearia sylvestris) against oral pathogenic microorganisms. The study will have three major research steps: 1- Evaluation of antimicrobial (antibiotic and antifungal) potential of crude extracts and fractions from plant species via minimum inhibitory concentration (MIC) and minimum bactericidal and fungicidal concentration (MBC/MFC) tests; 2- Assessment of the antibiofilm effect of extracts and fractions from plant species by determining the biomass via violet crystal method and the microbial population via platting of treated biofilms; 3- Evaluation of the inhibition capability of extracts and fractions from plant species on the initial steps of biofilm formation; specifically, the effect of treatments on the glucan synthesis on saliva-coated hydroxyapatite surface and on the microbial adherence to the salivary pellicle and to the initial matrix (glucans) on formed hydroxyapatite surface.
