Physiological and molecular responses of three models of running training with equalized loads in wistar rats

Abstract

Studies have shown improvement in oxidative activity and performance from different models of training. However, few studies analyzed the physiological responses from the manipulation of the variables volume and intensity with equalization of loads. The aim of this study is to verify the physiological and molecular responses of Wistar rats submitted to three acute training sessions, with equivalent loads. Seventy-eight rats will be subjected to training sessions, that will be: GZ1 (n=24), continuous running in intensity below the AnT (volume=43min, intensity=70% AnT); GZ2 (n=24) one running will be held at AnT intensity (volume=31min, AnT intensity=100%) and GZ3 (n=24), efforts supra-AnT (volume=25min, intensity=120.4% AnT). Before and after acute sessions and after 6, 12 and 24 hours, 6 animals in each group will be sacrificed for quantification of muscle content (gastrocnemius and soleus) of glycogen, glucose transporter (GLUT-4), proliferator-activated receptor- preroxisome y coativator 1-± (PGC1-±), AMP-activated protein kinase (AMPK) and citrate synthase (CS) by means of immunoprecipitation and immunoblot techniques. Then forty-two rats will be used to test the behavior of these same parameters in two models of polarized training using the previously standardized training sessions. For comparison between groups, at different times after training the One-way analysis of variance test (ANOVA) and Tukey post-hoc test will be used, with p<0.05. (AU)
