Abstract

Obesity is a chronic disease that affects millions of individuals and is associated with several metabolic changes. The interactions between genes and environment have been studied to identify possible factors that influence the responses of individuals to different metabolic conditions. The hypothesis of this study considers that severely obese patients have alterations in the genetic profile that would make them susceptible to this disease. Objectives - To characterize the genetic profile [analysis of up to 5 million Single Nucleotide Polymorphism (SNP)] and associate with biochemical variables, anthropometry, body composition in patients with grade III obesity. Methods - 25 individuals will be selected, divided into 2 groups: Intervention (G1) - 15 patients with morbid obesity [body mass index (BMI) e 40 kg/m2] before and after dietary intervention for 6 weeks; Control Group (G2) - 10 healthy normal-weight individuals (BMI between 18.5 and 24.9kg/m2). All subjects will be submitted to the collection of peripheral blood sample for analysis of the genetic profile (BeadChip-Illumina® 5 million SNPs) and biochemical, and anthropometric and body composition. Statistical analysis of the data will be used normality test (Shapiro-Wilk), descriptive statistics (mean and standard deviation), T test (paired and independent), Spearman correlation, and specific software for genetic analysis (Hierarchical cluster). It will be admitted level of significance set at p <0.05. (AU)
