Investigation of chalcones against Staphylococcus aureus MSSA and MRSA: evaluation of antibacterial, antibiofilm, anti-adsion activity and toxicity on Galleria mellonella

Abstract

Staphylococcus aureus is a commensal microorganism and human pathogenic microorganism. Which is responsible by variety of diseases, incluing acne, endocarditis, boils, osteomyelitis, pustules, impetigo, meningitis, pneumonia and arthritis. Several bacterial agents have developed resistance. In the scientific initiation project developed by the candidate, conducted by two Fapesp processes (2014/18577-5 and 2016/08084-7), we identified the activity against MSSA of three chalcones. In the present project, we propos to prepare these chalcones by the method established by the candidate during the scientific initiation period. The chalcones antimicrobial potential will be tested by biological assays, which will allow significant advances. These trials include assessments; A) antibacterial activity against MSSA and MRSA; B) inhibition of MSSA and MRSA biofilm production; C) effects on preformed MSSA and MRSA biofilms; D) inhibition of adhesion of MSSA and MRSA on human keratinocytes (HaCat cells); E) toxicity against Galleria mellonella, wich was selected as an alternative eukaryotic model; F) antibacterial activity against S. aureus strains resistant to macrolides and tetracyclines; G) antibacterial activity against other Gram-positive and Gram-negative bacterial species. The conclusion of the proposed project at Masters level will conduce the patenting and publication of the data developed by the candidate since the scientific initiation. (AU)
