Abstract The functionalization of metal surfaces by bioactive molecules has been investigated in order to promote osseointegration of implants in sites with poor bone quality in the jaws. Despite some evidences of biological benefits, the effects of functionalization of biomaterials with BMP-7 on the bone repair are still contradictory, and those of blood clot formation on the osteoblast differentiation induced by BMP-7- functionalized nanostructured titanium (Nano-Ti) should still be clarified. Thus, this project aims to: 1) evaluate the effects of supplementation of BMP-7 to the culture medium on the osteoblastic cell signalling and on the acquisition of the osteogenic phenotype, aiming to define the concentration and the exposure time interval of BMP-7 with the most significant biological effects; 2) evaluate ex vivo the effects of blood clot formation on the signalling and differentiation of osteogenic cells induced by the release, by simple adsorption, of BMP-7 on the Nano-Ti surface. In a mouse model, the blood clot will be formed on Nano-Ti discs functionalized with BMP-7 prior to MC3T3-E1 cell plating and culturing for periods of up to 21 days. The following parameters of the acquisition of the osteogenic phenotype will be evaluated: 1) cell morphology and proliferation; 2) expression of Runx2 and Smads 1, 5 and 9; 3) formation of mineralized matrix.
