Effect of topic PTH incremented to Biogran in alveolar bone healing of orchiectomized rats

Abstract

Osteoporosis is a systemic disease characterized by lower bone mass and deterioration of bone tissue microarchitecture. Studies show that osteoporosis and fractures by fragility are increasing even in women as in men. Drugs like PTH 1-34 are indicated for stronger osteoporosis once this is a anabolic agent of bone tissue, increasing the bone mineral density Therefore, the purpose of this project will be to evaluate a synthetic bone graft (Biogran) in association to PTH 1-34 in the alveolar bone healing of rats submitted to orchiectomy. 96 rats will be divided in two groups (Group I, Group II), according to the analysis that will be submitted. Into these groups, rats will be divided in two sub-groups (SHAM, ORQ), according to the surgery of osteoporosis induction. SHAM- fictional surgery, ORQ bilateral orchiectomy, and after this, they will be divided in 3 other groups: clot (alveolar defect without biomaterial), BG (alveolar defect filled with Biogran), BG-PTH (alveolar defect filled with Biogran PTH topical). Euthanasia will be performed at 60 days after tooth extraction. It will be performed microtomographycal, histological and immunohistochemical analysis in order to characterize bone remodeling (osteoprotegerin, osteocalcin, RANKL TRAP) as well as Wet pathway. All of the data will be submitted to normality curve to determinate the more appropriate test (parametrical versus non parametrical). Significance level of 0,05 all be used for all tests. (AU)
