Abstract Incorporation into cell membranes is key for the action(of photosensitizers in photomedicine treatments, with hydroperoxidation as the prominent pathway of lipid oxidation. Herein we shall fabricate Langmuir monolayers of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) and 1,2-dipalmitoyl-sn-glycero-3-phosphorylcholine as cellular membrane models to investigate the adsorption of the photosensitizer Eosin Y and the photo-induced effects of lipid oxidation. The Langmuir monolayers will be made onto ultrapure water subphase with different concentrations of Eosin Y (mmol/L -µmol/L), characterized by surface pressure (mN/m) vs. mean molecular area (À-A isotherms) measurements. The lipid membrane loaded with eosin will be further photo-activated to analyzed the effects of lipid oxidation on the membrane properties, which includes the increase of mean molecular area of the lipids and the cleavage of the carbonic tails.
