Influence of diabetes on pulp tissue mineralization of Wistar rats submitted to dental bleaching

In previous studies, dental bleaching with hydrogen peroxide (H2O2) is associated with inflammation or necrosis in the pulp tissue, subsequent formation of tertiary dentin, and osteocalcin (OCN) increase. In addition, we observed that the presence of diabetes mellitus may influence inflammation and formation of tertiary dentin in the pulp tissue of bleached rat's molar. Objective: this study aims to evaluate the influence of diabetes melittus on the dentinogenesis of pulp tissue of molars of Wistar rats submitted to dental bleaching, through the immunohisto chemical of OCN. Methods: Twenty-eight rats will be divided into 2 groups (n = 14): normoglycemic and diabetic rats. Diabetes mellitus will be induced by streptozotocin, diluted in citrate buffer, by intramuscular injection. After confirmation of diabetes, the rats will be anesthetized and the teeth will be bleached using bleaching gel with 17.5% H2O2 in the upper right molars of each group, for 30 minutes. The molars on the left side will be used as control. Thus, after dental bleaching, we will have 4 groups of 14 hemimaxilae each: normoglycemic (N), N-bleached (NCla), diabetic (D), and D-bleached (DCla). After 2 and 30 days, the rats will be killed and the hemimaxilae processed for histological and immunohisto chemical analysis for OCN. Scores will be assigned to the inflammation and immunolabeling, and submitted to the Kruskal-Wallis and Dunn statistical tests (P<.05). (AU)
