Abstract

This project aims to investigate different cultivation conditions of five filamentous fungi in order to evaluate the influence of the culture medium and the incubation time on the biotransformation reactions of ent-hardwickiic acid. It is also intended to select one or two filamentous fungi to biotransform this diterpene acid. The ent-hardwickiic acid is the major constituent of the Copaifera pubiflora species and is also present in other Copaifera oleoresins, which are marketed worldwide, and in Brazil, are the raw material of vegetable origin most commonly used in folk medicine. Biotransformation studies can bring knowledge about the metabolism of this substance, besides providing a way to obtain new derivatives. Filamentous fungi have an enzymatic apparatus similar to those of mammals, contributing to the elucidation of metabolic routes and to the knowledge about the formation of pharmacologically active, inactive or even toxic products for the organism. In order to reach the proposed objectives, five filamentous fungi will be cultivated under different conditions, extracts will be obtained from the culture broths and mycelia and all the extracts will be submitted to the chromatographic analysis using thin-layer chromatography and also high-performance liquid chromatography coupled to DAD and Corona CAD detectors.
