Novel bioinspired bonding agents for improving longevity of dental restorations

Abstract

The high incidence of replacement of resins restorations highlights the need for improved restorative materials. Objectives: synthesis and characterization of novel Epigallocatechin-galate (EGCG) methacrylate resins to inhibit bacterial colonization and proteins degradation. This approach will enable the modified ECGC to photopolymerize, creating resins with adequate mechanical properties and also antibacterial and antiproteolytic activities for the restoration life-span. Methods: EGCG will be modified by reaction with 1/3, 2/3 and 1 molar equivalents of methacyloyl chloride to introduce photopolymerizable double bonds. Monomer structures will be characterized by Fourier Transform Infrared Spectroscopy (FT-IR) and Nuclear Magnetic Resonance (NMR). The materials cytotoxicity to odontoblastic-like cells will be evaluated by a Methyl Tetrazolium (MTT) colorimetric (MTT) assay. Degree of conversion will be determined by FT-IR. Flexural Strength (FS) and modulus of elasticity (E) will be obtained using a universal testing machine. Antibacterial efficacy will be tested against S. mutans. Bacterial viability, cell adhesion and biofilm formation assays will be determined by CFUs and scanning electron microscopy. Antiproteolytic activity will be measured by a collagenolytic/genatinolytic enzyme activity, gelatin zymography, total protein and solubilized telopeptide assays. Antibacterial and antiproteolytic properties will be evaluated at two time-points: baseline and after 6 month ageing of polymerized resins in artificial saliva. One-way ANOVA and Tukey post hoc tests will be used to analyze cytotoxicity and mechanical properties. Remaining properties will be analyzed by Two-way ANOVA and Tukey's post hoc test (±=5%). (AU)
